Basically I'm struggling to evaluate on of my sites, and don't really know where to start.

The site: Webmaster Niche

PR3
Alexa 40k
Uniques p/m - 20k+
Page views p/m - 250k
Inbound links - 12k
age - 5 months
Revenue - just setup BuySellAds 3 days ago so assume none.

Any help or thoughts would be great!

It's tough to give a decent estimate if you won't disclose the URL, and the fact that the site has no revenue doesn't help either. Can you at least tell us:

What is the topic of the site?
Is it a forum, a blog, or something else?
What is the physical location of the visitors (what country are they in)?
How much time is required to manage the site?
Are there any expenses?

One more question: How much of profit would a buyer be able to reasonably see himself making from this site in the next 12 months if he had all the facts?

How much time does this take per day and per week, how many posts are there per day and per week, and what is the average word count? Blogs are a pain to manage because they die off without regular updates, so I either need to plan to spend the same amount of time you do, or I need to know how much it will cost to outsource the work.

Also, where is the traffic coming from? Are your visitors promoting the site, do you submit your posts to social networking sites, or are you just relying on search engine traffic?

profit I have no idea I'm not one for monetizing usually!

Blog posts are daily, some take 10 mins some take an hour, just depends on the detail I go into.

Traffic is from other design blogs, twitter, social bookmarks, and a little organic

I'd say that a fairly conservative estimate for the value of the site is $1,500, but it's hard to know without seeing the site or the domain.

The only way to know for sure is to put it up for auction.

I agree with benitez17, what we are all trying to do here is guess what buyers would value the site at, and they're going to base their valuations on how much they think they can earn.

It's next nigh impossible to guess the value some unknown third party is going to make based on his own guess about some unknown site.
